Charles Horace Mayo (July 19, 1865 - May 26, 1939) was an American medical practitioner and was one of the founders of the Mayo Clinic along with his brother William James Mayo, Augustus Stinchfield, Christopher Graham, Edward Star Judd Jr., Henry Stanley Plummer, Melvin Millet, and Donald Balfour .

Charles Mayo studied in the public schools of Rochester, and received the degree of Doctor of Medicine from Northwestern University in 1888. When Mayo had been home for about a year, assisting his father and elder brother in their practice of general medicine, the sisters of St. Francis founded St. Mary's Hospital in Rochester.
